Job Description Summary:

The Associate Scientist I, Gene Therapy is responsible for conducting scientific experiments that contribute to research and drug discovery activities at PTC. This position assists in production, purification, and characterization of recombinant adeno-associated viral (AAV) vectors to support in vivo efficacy and safety study in animals, as well as in the optimization of current AAV production methodology. The incumbent is responsible for communicating experimental results to his/her supervisor, the project team and for maintaining the AAV Database.

The Associate Scientist I, Gene Therapy supports adherence to all regulatory requirements and company Standard Operating Procedures (SOPs) including as appropriate.

Job Description:


Primary duties/responsibilities:
  • Under limited guidance, executes experiments and communicates results as part of multi-disciplinary team(s).
  • Applies relevant scientific principles and techniques to research problems.
  • Maintains detailed laboratory notebooks.
  • Maintains knowledge of state-of-the-art principles and techniques and may contribute to the identification of new project areas or technologies.
  • Ability to trouble shoot experiments -designing controls, generating interpretable data.
  • May provide technical and scientific input to the project team or research.
  • Presents findings internally and contributes to the publication of the work of the project team
  • Contributes to the development of patent applications as appropriate.
  • Complies with all safety requirements.
  • Involved in general lab duties as assigned, such as.
-Equipment maintenance.

-Identification of capital equipment needs

-Maintenance of clean and uncluttered work areas

-Maintain inventory and ordering lab supplies as needed.

Specific duties may also include, but may not be limited to, the following:
  • Performs production of rAAV vectors for in vivo efficacy and pharmacology studies.
  • Provides support to the team in the characterization of the produced rAAV vectors, including silver staining for purity, ELISA for capsid copies and serotype, transduction & Western blotting for MOI, alkaline gels, NGS sequencing and SEC-MALS for titer and full/empty ratio.
  • Supports development of new methods and optimization of the existing methods for AAV production in-house.
  • Maintains database of AAV and plasmid vectors.
  • Provides support for in vivo studies as required.


* Minimum level of education and years of relevant work experience.
  • Bachelor's degree in Biology with a minimum of 3 years' experience, or Master's degree in Biology with a minimum of 0-2 years of relevant experience.

* Special knowledge or skills needed and/or licenses or certificates required.
  • Excellent aseptic techniques with mammalian cell culture maintenance and cryopreservation experience.
  • In depth knowledge of Cell Biology and Molecular Biology. Knowledge of and ability to use standard molecular and cell biology techniques including DNA and RNA purification, quantification and transfection, molecular cloning, PCR, RT-PCR, qPCR, ELISA and Western Blot.
  • Ability to work with LIMS and databases.
  • Ability to independently execute experimental assays.
  • Candidate is expected to work comfortably in a fast-paced, collaborative team environment; be flexible and motivated; demonstrate excellent organizational skills, maintain full records of laboratory work and present results of studies at internal meetings.
  • Knowledge of bioinformatics is a strong plus.
  • Demonstrates technical proficiency, scientific creativity, and the ability to collaborate with others on cross functional teams in a fast-paced, matrixed, environment consisting of internal and external team members.
  • Possesses knowledge of scientific principles and concepts and keeps current with relevant scientific literature.
  • Proficiency with Microsoft Office- preferably MS Excel and MS PowerPoint.
  • Effective verbal and written communication and skills.
  • Analytical thinker with problem-solving skills and the ability to adapt to changing priorities and deadlines.
  • Effective planning, organization and time management skills including the ability to support and prioritize multiple projects.

*Special knowledge or skills and/or licenses or certificates preferred.
  • Knowledge of AAV methodology highly desired.
  • Working knowledge of NGS sequencing is a plus.
  • Ability to work with LIMS and databases.
