Occupational Summary

Perform a variety of complex technical duties involved in conducting high-throughput genomics experiments to support research efforts in the Reddy Lab at Duke University. The work performed will be part of a large-scale collaborative project involving the Ciofani, Gersbach and Crawford labs also at Duke. The successful applicant will be under the direction and supervision of the postdoc leading the project and will report primarily to him. He/she will be given ample opportunities to further develop his/her independent research skills. Details of the Reddy Lab can be viewed at http://reddylab.org

 

Work Performed

 

Major tasks to be performed will include: the management of a mouse colony through breeding and genotyping; mouse handling and dissections to isolate primary cells; large scale primary culture using sterile technique; expansion and differentiation of cells in preparation of experiments; transfection of cells using both transient and viral approaches; characterization and purification of cells using magnetic and fluorescence assisted cell sorting; targeted genome and epigenome editing using CRISPR/Cas9 based tools; isolation of nucleic acids through chromatin immunoprecipitation, RNA and DNA purifications; generation of high-throughput sequencing libraries for genome wide analysis. Additional tasks relating to molecular biology will include growth and preparation of electrocompetent bacteria; Gibson Assembly and plasmid cloning, transformation, and isolation of plasmid DNA from E. coli.

 

Perform routine laboratory tasks related to the above goals including preparation of reagent stocks, cell growth media, and buffers; ordering and stocking sufficient supplies and equipment for performing experiments; cleaning and maintain laboratory equipment.

 

Review results and quality control from completed experiments; record results in lab notebooks; communicate results in weekly update meetings with project leads. Option to contribute in weekly journal clubs and group meetings.

 

Perform other tasks and special independent projects as assigned.

 

Education/Training

 

Work requires a bachelor's degree in biology, biochemistry, molecular biology, genetics, or other science related scientific field.

 

Experience

 

Previous work handling rodents or live animals is an asset

Previous molecular biology or biochemistry experience is a plus.

Or an equivalent combination of relevant education and/or experience.
