Bacterial Vaccines Associate Scientist

The Bacterial Vaccines Associate Scientist will set up, operate, and maintain laboratory instruments, perform in vitro experiments, analyze data, records results, and provide interpretations and conclusions as appropriate. They will prepare bacterial and tissue culture growth media, prepare bacterial isolates and tissue culture cell lines for cryogenic storage, and will propagate cultures from cryogenic storage. 

The scientist will work within the Gene Therapy and Gene Delivery team to help develop novel Gene Therapies for unmet medical needs. The scientist will help develop Adeno-Associated Virus (AAV) producer cell lines and characterize other scalable AAV manufacturing technologies to support manufacturing for Gene Therapy programs in retinal, neuro, and infectious diseases. We are seeking someone who excels in a group environment, can work collaboratively on cross-functional teams, and has a willingness to handle increasing levels of responsibility. 

Primary duties will include the qualification and performance of assays associated with full development, discovery and early development programs including ELISA and Opsonophagocytic Killing (OPK) assays, and serological assays for the characterization of bacteria. The candidate will handle human cell lines, human blood and blood products, isolate immune cells from human blood, and manipulate these cells in bacterial and tissue culture assays.  They will be expected to perform or assist in the performance of in vitro assays in support of ongoing clinical trials under GCP and non-GCP conditions. Duties will also include the aseptic growth and maintenance of continuously propagated tissue culture cell lines, the analysis of cell titers, and the assessment of cell differentiation using flow cytometry and live-dead staining techniques. 

Key Responsibilities:
Daily maintenance of mammalian cell cultures,
Transfection and/or electroporation and selection of stable clones/ pools,
Expansion and screening of clonal cell lines,
AAV and Adenovirus production, and screening clones for biological activities and/or virus production. 
Candidates should have a working knowledge of viral-based production technologies and quality control (QC) techniques, viral titration assays (qPCR/ddPCR) as well as analytical assays such as silver stain/western blotting, fluorescence microscopy, luciferase, and SEAP assays.

Experience and Skills Required:
BS/BA degree is required
Basic microbiology lab skills, ambitious and willing to learn.
Willing to train recent grads with lab experience. 
Prefer tissue-culture and aseptic technique 
Experience generating and characterizing stable cell lines.
Strong critical, scientific, and strategic thinking skills allowing individual to work independently with minimal supervision

A general understanding of virology and cell biology vectors is preferred.
Knowledge of viral vector purification is preferred
A broad understanding of viral titration assays (qPCR/ddPCR), silver staining/western blotting, DLS/DSF, ELISA, FFF-MALS, HPLC, and confocal imaging.
Experience with documentation and reporting of results using electronic lab notebooks is preferred
