We are currently recruiting a Research Technician III for a collaborative project in lab to support a study focused on pre-clinical validation of gene therapy approach for Parkinson’s disease based on viral platforms and CRISPR/Cas technology.


Essential Duties and Responsibilities

This position will heavily rely on the expertise in animals (mice) work. The prospective candidate must be proficient with basic mouse handling procedures, husbandry, colony maintenance. Expertise and proficiency in stereotaxic injections ishighly preferred. Proficiency and extensive experience in basic molecularbiology and cell biology techniques including assay development skills are required for this position. That includes immunoblotting, flow, IHC, histology and pathology analyses and real-time PCR. Previous experience in work with viral vectors is a plus, but not mandatory. The prospective candidate will have good communication skills, allowing effective communication with other team members and the PI. The person will establish work flow with the help of principle investigator and core staffand expected to learn and execute procedures and experiments independently with minimal supervision. The person will be expected to supervise and help the work of lower level employees, such as students and to train new personnel.


The person will manage the lab’s projects related to PD-AD animal models.

(20%): Perform a variety of complex and independent activities involved in the collection, analysis, documentation and interpretation of neuroscience-related work  
- Independent data collection, management, and analysis of all mouse behavioral data for Cortes group 
- Interim coverage of mouse management for lab 
- Independent data collection, management, and analysis of multiple molecular biology techniques (immunoblotting, invivosurgeries, histology) 
- Management of lab supply inventory 
- Management ofmouse colony comprised of six different transgenic lines, including breeding, genotyping, euthanasia, experimental cohort set ups 
- Generation of large cohorts of double transgenic and homozygous lines required for the data collection described above
(20%): Establish work procedures; select sources of information and specific data; setup interviews with appropriate authorities or subjects and prepare correspondence to locate the specific data required 
- Research on existing protocols and published literature to select appropriate reagents/materials for use on specific projects, including independent research and troubleshooting of new research topics 
- Writing new protocols for behavioral testing, western blotting, immunohistochemistry, animal dissection and animal survival surgery 
- Development of new dissection methods for varied tissue preparations 
- Establishing new staining and immunoblotting techniques 
- Communication with internal Duke Core groups (Genomics Core, Neurobehavioral Core) and external collaborators (Barhop Institute) to coordinate testing, data acquisition, or animal transfer 
- Coordination with other researchers and managers in the Center to plan upcoming collaborations/experiments


(15%): Oversee behavioral evaluation of mice for motor and neurological function and cognition / anxiety 
- Perform or acquire training to perform minor surgical procedures on mice, including intraperitoneal, stereotactic brain and tail-vein injections. 
-Acquire training to perform culture of  primary neurons and primary myoblasts 
- Prepare tissue and specimens from mice for complex procedures, including microscopy and immunoblotting, and devise experimental techniques for tests and analyses.


(15%): Evaluate and interpret collected data and prepare oral presentations or written reports and analyses setting forth progress, adverse trends and appropriate recommendations or conclusions 
- Write-up/oral presentation of current data and project progress to group of peers and Supervisors


(10%): Develop forms and questionnaires and coordinate the application of research techniques 
- Production and maintenance of data sheets for ongoing and future experiments 
- Planning and coordination of longitudinal mouse projects requiring multiple stages of behavioral or biochemical testing


(10%): Confer with principal investigator in developing plans for research projects and to discuss the interpretation of results and the preparation of manuscripts for publication 
- Regular meetings with PI to report project progress, relay relevant data/concerns, and strategize for future  experiments
- Data collection, manuscript writing
- Coordination with Ce nter mouse colony manager to ensure organization and health of colony


(8%): Supervise the work of lower level employees engaged in the collection and tabulation of data; train new personnel 
- Direct supervision and training of 3 undergraduate researchers on mouse handling and molecular biology techniques under Dr. Chiba-Falek 
-Supervision and training of  lab employees and students on mouse phenotyping and surgical techniques (as needed) 
- Management and delegation of mouse colony maintenance


(2%): Perform other related duties incidental to the work described herein


Minimum Qualifications

Work generally requires skills acquired through the completion of a bachelor's degree in botany, biology, zoology, psychology or other science related scientific field program. Strongly prefer BS degree or equivalent in medical sciences, genetic or molecular biology.


Work requires two years of research experience. A related master's OR AN EQUIVALENT COMBINATION OF RELEVANT EDUCATION AND/OR EXPERIENCE degree may offset required years of experience on a 1:1 basis, e.g., a two year master's degree in lieu of two years of experience.


Salary would commensurate with previous experience and background.
